小分子的三维结构

单晶X射线衍射最常见的获得详细分子结构的实验方法可以解析单个原子,单晶X射线衍射(SXRD)的执行是通过分析许多相同分子(单晶)的规则序列的X射线衍射图进行的。许多纯物质,从小分子到有机金属复合物、蛋白质和聚合物,可以在适当的条件下凝固成晶体。当凝固成晶体状态时,这些单个分子通常适应仅有的几个可能的三维方向之一。当单色X射线通过一个单晶时,射线与原子中的电子相互作用,从而产生一种射线散射形成一个独特的图像。当晶体在X射线中旋转时,多个图像通过使用一个区域X射线检测器被记录。通过计算这些图像的强化分析,可以分析小分子的三维结构。
